John Chen received his PhD in Biophysics/Cell Biology at the Scripps Research Institute in 2015. While in the lab of R. Luke Wiseman, Dr. Chen's graduate work focused on the impact of endoplasmic reticulum (ER) proteostasis activity on the secretion and quality of secreted proteins related to systemic amyloid diseases.
Currently, Dr. Chen is interested in tauopathies—a class of neurodegenerative diseases associated with the proteotoxic aggregation of tau. He is interested in understanding the molecular mechanisms that dictate tau aggregation as well as its intercellular propagation. Because tau aggregates have unique prion-like conformations that have unique cellular targets, he is also interested in identifying genes that contribute toward this proteotoxic selectivity. As such, Dr. Chen uses a high throughput CRISPRi/a library screen in a fluorescent reporter cell line to identify genes associated with tau proteostasis.
